Pregabalin is an odorless white crystalline powder chemical. Chemical name 5-ethyl-5-phenyl-dihydro-4,6(1H,5H)-pyrimidinedione, molecular formula is C12H14N2O2, molecular weight is 325.4242, melting point is 281~282℃, slightly bitter taste, no acidity, in Slightly soluble in ethanol, almost insoluble in water, acetone or benzene.

The production of Pregabalin requires adding ethyl acetate into sodium ethoxide, refluxing for half an hour, cooling to room temperature, adding diethyl ethyl phenyl malonate and thiourea, and adding flow for 9 hours. Cool to 0°C, add ethanol, and dropwise add hydrochloric acid at 0-5°C to pH=3-3.5. Add zinc powder, heat up to reflux, slowly add hydrochloric acid, keep the reaction solution with 2-3% acid content, after adding, keep warm for 3h. Filter while hot, and recover ethanol from the filtrate. Cool to 20°C, add water, filter, and wash the filter cake to obtain Pregabalin.
1. Indications for Pregabalin
The effect is similar to that of phenobarbital. About 25% of it is oxidized to phenobarbital in the body, and the other part is cleaved to phenethylmalonamide. Primidone and its metabolites have antiepileptic activity. Other antiepileptic drugs are ineffective for grand mal seizures, psychomotor seizures, and petit mal seizures. Combined use with phenytoin can enhance the efficacy.
Dosage and usage of Pregabalin
Oral: start with 0.15g each time, gradually increase to 0.2g each time, 3 times a day. The maximum amount is 2g a day. Children 12.5 to 25 mg per kilogram of body weight per day, divided into 2 to 3 times.

1) The common adverse reaction is vomiting, so it is advisable to start with a small dose and gradually increase it. There are also symptoms such as lethargy, ataxia, and occasionally giant cell anemia.
2) People with insufficiency of liver and kidney should not use it.
3) Pregnant women are prohibited. Not to be used in combination with phenobarbital. Severe hepatic and renal insufficiency is contraindicated.
Pregabalin can be used to treat grand mal seizures, simple partial seizures, and complex partial seizures, but it is not a first-line drug. It should only be considered when phenytoin sodium, phenobarbital, and carbamazepine are ineffective alone or in combination. Pregabalin is more effective if combined with phenytoin sodium. Deoxyphenobarbital is less effective for petit mal, but effective for myoclonus in young children.
